Regulation of glycolysis occurs through 3 enzymes - hexokinase, PFK, and pyruvate kinase. Hexokinase is regulated by substrate-level regulation. PFK is regulated allosterically by ATP (turns off), AMP (turns on), citrate (turns off), and F2,6BP (strongly turns on). Note that ATP and citrate will be present when cells have a lot of energy, so it is logical that they turn off the enzyme. By contrast, AMP is present when cells have little energy and it is logical that it turn on the enzyme under these conditions. F2,6BP can counteract the negative effects of ATP on the enzyme to some extent. 2. Note also that PFK is very unusual in being negatively regulated by a molecule (ATP) that is also a substrate. This is possible because the enzyme has an allosteric binding site for ATP in addition to the substrate binding site and the Km for the allosteric site is higher than the substrate binding site. 3. Pyruvate kinase is regulated both allosterically andby covalent modification (phosphorylation/dephosphorylation). Phosphorylation of the enzyme by a protein kinase turns the enzyme activity down, whereas F1,6BP acts as an allosteric activator. This activation is known as feedforward activation. 4. Supreme Court and American gun ownership The Supreme Court, or at least 5 of the 9 justices, upheld the second amendment to the USA Constitution, which reads, "A well regulated Militia, being necessary to the security of a free State, the right of the people to keep and bear Arms, shall not be infringed." Those who want to restrict American access to hand guns read the above amendment as stating guns are only necessary for a "well regulated Militia." Meaning for a state's National Guard unit. Since weapons are provided by the Guard, there is no reason for the populace to own a hand gun. The advocates for gun ownership read that amendment as indicting it is the right of all Americans to own a gun, "a well regulated Militia," not withstanding. So goes the debate, which the Supreme Court seems to have settled in its recent decision. The best way to settle this debate is to look to the very men who wrote the 2nd Amendment to the United States of America Constitution. Now understand most of the 55 delegates to the United States Constitution Convention, went on to serve in government in various positions from town councils, mayors, state assemblies, governors, congress, senate, and the office of president. The question to ask is, once the Constitution was ratified and these men ascended to various public offices, why did they not confiscate all the pistols in these here United States? Lindy Hop and Charleston Routine about phosducin for "Dance Your PhD 2009" "Dance your PhD 2009" contest entry Rachelle Gaudet Michael Richters (aka transducin beta-gamma) and I (aka phosducin) use Lindy Hop and Charleston to illustrate my PhD thesis: Structural ***ysis of phosducin and its phosphorylation-regulated interaction with transducin beta-gamma. These proteins function in our eyes, as part of the retinal proteins that transmit visual signals to the brain. In a bright environment, phosducin binds to transducin beta-gamma and the two are found in the cytoplasm. Upon dark-adaptation, phosducin becomes phosphorylated, while transducin beta-gamma returns to the cellular membranes to increase the responses to small amounts of light. During my PhD, I determined the crystal structure of the complex of phosducin and transducin beta-gamma. In this complex, phosducin's N-terminal half interacts extensively with beta-gamma, illustrated in our Lindy Hop dance, a partner dance which relies on intricate lead-follow technique. We dance the Lindy Hop under bright lights in the back of the room, because the complex phosducin/transducin beta-gamma forms when our eyes are adapted to bright light. When eyes adapt to the dark (illustrated by our transition to the darkened front of the room), phosducin is released from beta-gamma and becomes phosphorylated (represented by the hat).
