Hoh, Ph.D. Johns Hopkins School of Medicine In multicellular organisms cells have highly specialized functions that depend on signals from their local microenvironment. Understanding the relationship between these signals and the cellular responses requires the ability to quantify them. In many simple cases there are direct ways for doing so, such as measuring the concentration of a ligand or the steepness of a gradient. However, important signals are often compositionally and spatially complex. For example, in a developing nervous system axonal growth is directed by the composition and distribution of molecules in the extracellular environment. Likewise, the migration of cancer cells involves responses to the organization of specific molecules in the surroundings. One powerful approach to understanding how cells process spatial signals is based on using micropatterned substrates to control the distribution of signaling molecules. Here we propose that the Shannon information theory formalism provides a robust and useful way to quantify the organization of proteins in micropatterned systems. To demonstrate the use of informational entropy as a metric we microfluidically patterned of lines of fibronectin with varying information content. Mary Beckerle (Univ Utah) Part 1: Adhesion, Signaling and Cancer Cell-substratum adhesion is mediated by integrins, a family of transmembrane, heterodimeric, extracellular matrix receptors that are concentrated at focal adhesions. Integin engagement influences a variety of signaling pathways and regulates cell behaviors including motility, proliferation, and survival. Disturbance of normal integrin function is associated with a variety of pathologic conditions including cancer. In the first segment of my seminar, I provide a broad overview of the cancer problem for a lay audience. Advances in our understanding of cancer as a genetic disease are discussed. 5. Cell Culture Engineering Frontiers of Biomedical Engineering (BENG 100) Professor Saltzman reviews the concept of gene therapy, and gives some examples of where this is applied. Methods to help deliver DNA into cells using viruses and cationic lipids are discussed, as a way to overcome some challenges in gene therapy. Next, Professor Saltzman gives a brief introduction into bacterial and mammalian cell physiology. He describes the different tissues in the body, the cell development/differentiation process, the anchorage dependence of mammalian cells that allows them to form an organism, and the extracellular matrix.
