Cellular Responses to Micropatterns of an Extracellular Matrix Protein with Defined Information Cont "Cellular Responses to Micropatterns of an Extracellular Matrix Protein with Defined Information Content" Jan H. Hoh, Ph.D. Johns Hopkins School of Medicine In multicellular organisms cells have highly specialized functions that depend on signals from their local microenvironment. Understanding the relationship between these signals and the cellular responses requires the ability to quantify them. In many simple cases there are direct ways for doing so, such as measuring the concentration of a ligand or the steepness of a gradient. However, important signals are often compositionally and spatially complex. For example, in a developing nervous system axonal growth is directed by the composition and distribution of molecules in the extracellular environment. Likewise, the migration of cancer cells involves responses to the organization of specific molecules in the surroundings. One powerful approach to understanding how cells process spatial signals is based on using micropatterned substrates to control the distribution of signaling molecules. Here we propose that the Shannon information theory formalism provides a robust and useful way to quantify the organization of proteins in micropatterned systems. To demonstrate the use of informational entropy as a metric we microfluidically patterned of lines of fibronectin with varying information content. Fibroblasts grown on these patterns were sensitive to very small changes in informational entropy (6.6 bits), and the responses ...
Mary Beckerle (Univ Utah) Part 1: Adhesion, Signaling and Cancer Cell-substratum adhesion is mediated by integrins, a family of transmembrane, heterodimeric, extracellular matrix receptors that are concentrated at focal adhesions. Integin engagement influences a variety of signaling pathways and regulates cell behaviors including motility, proliferation, and survival. Disturbance of normal integrin function is associated with a variety of pathologic conditions including cancer. In the first segment of my seminar, I provide a broad overview of the cancer problem for a lay audience. Advances in our understanding of cancer as a genetic disease are discussed. The influence of cell adhesion on control of cell growth is reviewed. See more at
5. Cell Culture Engineering Frontiers of Biomedical Engineering (BENG 100) Professor Saltzman reviews the concept of gene therapy, and gives some examples of where this is applied. Methods to help deliver DNA into cells using viruses and cationic lipids are discussed, as a way to overcome some challenges in gene therapy. Next, Professor Saltzman gives a brief introduction into bacterial and mammalian cell physiology. He describes the different tissues in the body, the cell development/differentiation process, the anchorage dependence of mammalian cells that allows them to form an organism, and the extracellular matrix. OLIgo Mass Profiling (OLIMP) of Extracellular Polysaccharides Watch video protocols on JoVE - . Journal of Visualized Experiments (JoVE) is a scientific journal publishing video articles on biological experiments filmed at leading research institutions. JoVE is the first video journal indexed in PubMed. To see more of this protocol go to OLIgo Mass Profiling (OLIMP) of Extracellular Polysaccharides This video protocol was filmed at the University of California-Berkeley. The direct contact of cells to the environment is mediated in many organisms by an extracellular matrix. One common aspect of extracellular matrices is that they contain complex sugar moieties in form of glycoproteins, proteoglycans, and/or polysaccharides. Examples include the extracellular matrix of humans and animal cells consisting mainly of fibrillar proteins and proteoglycans or the polysaccharide based cell walls of plants and fungi, and the proteoglycan/glycolipid based cell walls of bacteria. All these glycostructures play vital roles in cell-to-cell and cell-to-environment communication and signalling. An extraordinary complex example of an extracellular matrix is present in the walls of higher plant cells. Their wall is made almost entirely of sugars, up to 75% dry weight, and consists of the most abundant biopolymers present on this planet. Therefore, research is conducted how to utilize these materials best as a carbon-neutral renewable resource to replace petrochemicals derived from fossil fuel. The main challenge for fuel ...

Intro to Foregen - non-profit agency to promote foreskin regeneration Circumcision removes the best part of the penis. An emerging field of regenerative medicine is using "extracellular matrix" which when applied to an open wound seems to cause the body to figure what is missing and regrow lost parts. is an Italian non-profit (ONLUS) agency which aims to promote research into applying this technology to circumcised men. Hundreds of thousands of men are enduring a tedious multi-year tensioning process of non-surgical foreskin restoration to regrow enough penile skin so they can get back many foreskin functions. *** is invariably dramatically improved, however, these techniques can never replace the lost specialized pleasure-receptive structures of the ridged band and frenulum. Regeneration with extracellular matix could potentially give a man back these valuable parts. Featured in this video Vincenzo Aiello of Foregen and Ron Low of TLCTugger.
