Introduction to Microbiology Culture Techniques Nicole Gentile, PhD Candidate, provides an overview of basic microbiology and the concepts involved, including the bacterial growth curve and classifying organisms based on morphologies. This lecture describes blood, urine and skin/soft tissue cultures, focusing on the types of media, sample collection processes, culture procedures, as well as speciation and susceptibility testing. Basic staining procedures, such as the simple stain, gram stain, spore stain, negative stain, and acid fast stain are briefly discussed. Included in the staining procedure descriptions are explanations of the organisms that the stains identify. In addition to staining procedures, biochemical tests used for differentiating bacteria are covered. Concluding the lecture are some facts about fungi and viruses, focusing on the current 2009 novel H1N1 influenza pandemic.

When the cells in adherent cultures occupy all the available substrate and have no room left for expansion, or when the cells in suspension cultures exceed the capacity of the medium to support further growth, cell proliferation is greatly reduced or ceases entirely (see Figure 4.1 below). To keep them at an optimal density for continued growth and to stimulate further proliferation, the culture has to divided and fresh medium supplied. Kalash of Cultures - Pakistan Sep 2003 In a remote part of northern Pakistan, a tribe believed to be descended from Alexander the Great is facing cultural annihilation. As animists, the Kalash Kafirs live by cultural practices which clash with those of the Muslim Pakistanis' that surround them. Their identity is constantly undermined by their Muslim neighbours. Help comes in the form of volunteers from Greece who believe that the Kalash people are part of their own ancient history and worth preserving. New facilities are being built but such help comes at a price. "They are building in their own way...I think they should build like Kalash," complains villager Mohammed. Whether fact or fiction, the Kalasha realise the commercial value of their founding myth and so are keen to preserve it. However, it is their spiritual life and beliefs that they feel are under threat by the growing hostility of fundamentalists. With jobs promised to those that convert, over 6000 have reluctantly given up their bright national dress to become Muslim. For the Kalash people, both their past and future remain uncertain.
